using an audio interface and a DI box you could just record the input and output of the amp you want to test, level match the signals and then calculate A-B for the difference signal. Then you would know if there is anything possibly audibly different between the amps as you have created a 'best case' test with no masking sound.

if the input is pink noise band-limited to one octave, then all harmonic distortion and IM products will fall outside the octave which covers the input signal. This will make it much easier to see any type of distortion.

Absolutely!
The loudspeaker's impedance curve, at the given frequency, will play a factor, on how the amplifier will behave amplifying music signals. Not even two loudspeakers, offering the same brand/model number, will offer identical impedance curves, within a 20 Hz – 20 kHz range. This can be easily found using LIMP, the Woofer Tester and/or, Room EQ Wizard.
Overall, the amplifier needs to manage various behavioural patterns, under the given circumstances, all courtesy of the loudspeaker.
