You dont need a router just nice if you have one, i get acceptable results without one, Flat packs would just need sanding etc...
Money to be made in Flat pack G-subs fe sure Wink

50quid router, 5 quid roundover bit.

Not hard, Blimey I thought this was a DIY site.

In all honesty, a 40 quid battery driver, 50 quid router, and a jig saw will do for the speaker holes.
Just get the wood yard to cut up. The bracing does not need to be in any way as complicated as the plan. Do three cross braces across the back then one long one  the length sloted into the crosses. I've done this, it IS enough, doesn't rattle and is simple. Use metal bar handles, just use jigsaw carefully. You can recess later in life when the skills develop. Try one in MDF, B&Q prices mean you can try one for 30 quid. Make notes of your errors, then do the posh wood when you're ready.
I'm an impatient, inacurate woodworker, yet I have certainy never had a cab rattle or come apart.

Go for it. Slow, stop. think.
Use butt joints, they are fine. Make sure speaker holes are central and not too big, nice tight fit. Cut them before assembly. T Nuts can be done after build.
Handle holes: If using metal handles you can do them after build. If routing, do first, you need to get inside rounded as well! Think about handle position, do you need top hap, maybe feet? Bracing, dont forget it's there and try and add top hat or handles through it! Dont presume cheap ply is btter to work with than mDF, is not always, but should be more durable. Moisure resistant MDF is actually nice to work IMHO.
Dont forget to set front back enough for grills and excursion of driver, makesure you can find baton ofcorrect depth for grills. (B&Q do 25x25).
Have fun.
It just is.
