It's going to depend on the driver you've chosen.  If you've said what it is, I've missed it.  The stronger the motor and the cone, the higher the compression ratio you can use, up to a limit as previously stated.

You will find in the simulation that as you reduce the throat you'll start losing low extension so that may guide you.

For comparision, I've been playing with 8" mid horns and ended up with a compression ratio of just 2.5:1 and I reckon that's definitely as high as I'd want to take it.  In your case with a 10", that would make the throat no smaller than about 150cm2.

You can, but as several folks have said any gain is likely to be above the range you use it in and there's the risk of cancellations too.
If you use the offset driver option (double click on Nd to get a popup showing the different driver arrangements) you set S1 to as close to zero as possible, S2 to the area of the "horn" level with the centre of the drivers and S3 to the mouth area, it will give you the theoretical output.
Here's my rough estimate, obviously I don't know the exact dimensions of your box but scaling from your 10" horn I've figured your mouth would be about 45.5cm square(ish). My assumptions for length may be a bit optimistic, yours may be a little shorter (ooh-err!).
I've put in two drivers at 2V total input to keep it to the equivalent of 1W/1m for the pair, accounting for the apparently lower sensitivity. Black is vented, Grey is with the horn effect.
(Interestingly enough, you can see a similar effect in the response curves of the bigger EAW subwoofers that use this type of layout - you can see they have a small peak in their response matched by an increase in impedance at about 200Hz for the dual 18's and 130Hz for the dual 21.)
